We chose 3 established antioxidants, ascorbic acid, glutathione, melatonin to compare their protective effects in a DNA damage test by Comet assay. Methods: The target cells were human lymphocytes incubated with hydrogen peroxide to induce a baseline DNA injury. Results: We found that antioxidant property was dependent on the dose of the chemical used. At high concentrations, both dietary ascorbic acid and glutathione alone produced considerable DNA damage to the cells. This pro-oxidant effect was minimal when cells were treated with melatonin, a natural hormone released from the pineal gland. Conclusion: The importance of dose-response relationships is highlighted when investigating any beneficial antioxidant effects in vitro. Also, the concentration of antioxidant used in bench assays ought to be matched with prescribed dose for dietary supplementation, assuming that absorption is adequate. Methods: In this cross-sectional observational study, 60 eyes with POAG suspect suspicious disc, normal fields SAP ; and 25 eyes with early to moderate POAG glaucomatous disc changes and visual field defects on SAP and Matrix ; were evaluated on Matrix and SAP using the 24-2 strategy, and GDxVCC. Global indices MD, PSD ; were used for comparison of visual fields. TSNIT average thickness and NFI on GDxVCC were correlated with global indices of the perimetric tests. All eyes had visual acuity better than 6 9 with correction, refractive error of less than 5D sphere and 3D cylinder with no media opacities and normal pupils. Results: Mean age of patients was 55.6 7.3 yrs and 58.6 6.4 yrs in POAG suspect and POAG group respectively. In POAG suspect, mean MD and PSD by SAP vs. Matrix were -2.581.89 vs.-3.483.50 and 2.02 1.05 vs.3.280.75 respectively. There was a statistically significant difference in both MD and PSD of SAP and Matrix p 0.001, p 0.047 ; . In POAG group, mean MD and PSD by SAP vs. Matrix were -7.31 4.29 vs. -7.73 3.76 and 6.28 4.26 vs.5.65 2.63 respectively. In this group there was no significant difference in the MD and PSD using any of the two techniques of perimetry. Patients and Methods: Eighteen patients with cervical cancer were enrolled in this study. Cisplatin 30mg m 2 ; and paclitaxel starting dose 40mg m 2 ; 5 mg m2 escalation per level ; were given on day 1 of radiotherapy and then weekly for 6 cycles. Cohorts of 3 patients were enrolled at each level and 3 other patients were included if 1 or unacceptable toxic events UTE ; were recorded. Four levels were studied paclitaxel 40, 45, 50, mg m 2 ; with 3, 5, 4, and 6 patients enrolled, respectively. Radiotherapy was given to the pelvis with a fourfield box technique 5 days a week. Patients received 65 Gy in 1.8-Gy fractions. Results: All patients were evaluable for toxicity. The MTD was found to be paclitaxel 50 mg m 2 wk and cisplatin 30 mg m2 wk. Diarrhea was the dose-limiting toxicity. At level four, 3 patients showed UTE 2 grade 3 diarrhea and 1 grade 4 neutropenia associated with grade 3 diarrhea ; . Thirteen patients were evaluable for response: 7 complete and 5 partial responses were obtained with an overall response rate of 92.3%. Conclusions: Cisplatin 30 mg m2 wk and paclitaxel 50 mg m2 wk represent the MTD when they are administered concurrently with pelvic radiotherapy. Gastrointestinal toxicity is dose limiting. Concurrent chemoradiotherapy with paclitaxel and cisplatin is feasible; data suggest that this combination could be very active in patients with locally advanced carcinoma of the cervix.
